The sixth amino acid from the amino terminus of the β-chain of HbA is glutamic acid (a negatively charged amino acid). A mutation in one of the bases from an A to a T of the gene coding for glutamic acid in the DNA, changes the amino acid from glutamic acid to valine (no charge at neutral pH) at the same position. The α-chains of HbA and HbS are identical.
